Plasmid versus cloning vector as named roles are two labels for different claims. A plasmid is the DNA molecule — usually a circular, extra-chromosomal replicon. A cloning vector is the job of accepting an insert and keeping it in a host. The same file can wear both names only when it is doing that job. A naturally occurring plasmid can confer antibiotic resistance or carry metabolic genes without ever offering a usable multiple-cloning site, a documented copy-number regime, or a lab host that you actually transform. Calling that molecule a cloning vector smuggles in a job no one assigned. Exam tables that say “all plasmids can serve as cloning vectors” are teaching a convenience, not a definition.

Do not turn this into an insert-size law. Teaching pages disagree on how many kilobases a plasmid, cosmid, or BAC will stably hold. Those cutoffs are typical ranges for particular systems, not a reason to rename the molecule. If you need a size decision, you need the insert and the host, not a new synonym.

Not an Expression Cassette and Not a Second Replicon

You owe vector when you are claiming the carry-and-maintain job, or when you are distinguishing that job from phage or BAC vehicles sitting in the same freezer box.
