E. coli host-strain choice for a cloning vector is a genotype decision, not a broth contest. Pick the strain that keeps this insert: recA- and endA- for ordinary plasmids, a stability class when repeats or viral LTRs delete in the weekly cells, and dam-/dcm- only when a later cut is blocked by methylation. The richest medium does not decide that. Molecule versus cloning-job names and shuttle versus backbone are different pages. So is choosing the origin-plus-marker pair.

Thermo Fisher's cloning-strain note is explicit on one limit: dam-/dcm- cells should not be the routine transformation host, because those mutations raise the chance of unwanted changes in the propagated DNA. Use that class when a later ClaI- or XbaI-class cut is blocked by methylation. Do not use it because unmethylated DNA sounds cleaner. Prefer the routine class when the backbone is an ordinary cloning vector and the insert has no deletion history. Prefer the stability class when the same map comes back missing a repeat or an LTR. Prefer the methylation-free class for one unmethylated prep, then move the verified plasmid back to a recA- endA- host for storage. Name Strain Constraints Before Opening the Map
