Transient Transfection: Expression Without Stable Integration
Transient transfection is the introduction of nucleic acid into eukaryotic cells for short-term expression without a requirement that the DNA integrate into the host genome. The plasmid — or the RNA — works this week. It is not a genome edit. Mammalian workers mix the job with stable line generation and with knockout or knock-in language because all of those workflows start by getting nucleic acid into a cell. The start is shared. The claim is not.
The Plasmid Works This Week; It Is Not a Genome Edit
The category is a delivery-and-expression method. The purpose is a temporary change in what the cell expresses. The audience is anyone deciding whether this week's assay can run on extra-chromosomal DNA, or whether they must keep a selected clone. Kim and Eberwine (2010) draw the line that still holds in later reviews: stably transfected material is integrated and kept through replication; transiently transfected genes are expressed for a limited time and are not integrated. That is the definition. It is not a protocol for a lipid reagent.
Thermo Fisher's types-of-transfection page uses the same split: in transient transfection the nucleic acids exist in the cell for a limited period; in stable transfection they persist and are passed to progeny. Supercoiled plasmid DNA is described as the efficient DNA form for the transient job. RNA, including mRNA or siRNA, can also be transient. RNA that never enters the nucleus is still not a genome edit.
Why the Signal Fades in Days

The usual planning window is days, not weeks of antibiotic selection. Thermo's page, as captured in live search on 4 September 2026, says the transgene can often be detected for one to seven days and is typically harvested 24 to 96 hours after transfection, because nucleases and cell division remove or dilute the material. A 2021 methods overview says the same in review language: transiently transfected cells express the foreign gene without integration, so the new gene is not replicated as a chromosomal locus, and expression usually lasts several days before it is lost to division or other factors.
That is why a transient experiment is a calendar job. If the assay cannot be read inside the window, transient transfection is the wrong claim, even if the transfection itself "worked" on day one. mRNA can appear faster than plasmid DNA because it does not need nuclear entry. It is still transient. Dilution still wins if the cells keep dividing and you keep waiting.
Stable Transfection Is a Different Job
Stable transfection asks a subset of cells to integrate a cassette — usually with a selectable marker — and then spends weeks killing the rest. The output is a line, or a pool, that keeps the transgene through passage. That is a different job from this week's expression. It is also not automatically a knockout or a knock-in. Integration of an expression cassette is not the same claim as cutting a locus. Knockout versus knock-in owns those genome-edit jobs. Prefer transient transfection when you need protein, reporter, or knockdown this week and you can accept cell-to-cell variability and a fading signal. Prefer a genome-edit design when the scientific claim is about the locus, not about extra-chromosomal expression. A fluorescent burst on Thursday is not evidence that the genome changed.
